Bia2iTC (beta release) software for integrated use of Biacore & Microcal systems
Combining kinetic & thermodynamic characterization Kinetic characterization provides information about the rate of complex formation and its stability, whereas studies of interaction thermodynamics are required to understand why interactions occur. GE Healthcare supplies equipment for both label-free measurements of binding kinetics (Biacore™ instruments) and binding thermodynamics (MicroCal™ instruments). This provides a powerful combination of orthogonal techniques, bringing complementary information to the analysis and adding confidence to decision-making in key applications such as hit validation in drug discovery.
To facilitate the use of Biacore affinity data to optimize experimental design in complementary MicroCal assays, Bia2iTC (beta release) software is now available for free to Biacore system users (download requires a valid Biacore product key).

Overview & systems supported Affinity (KD) values determined using Biacore are imported into this software. The program uses the KD values to design all sample concentrations and run parameters needed to perform optimized ITC experiments. This data is subsequently exported as a setup file to the MicroCal instrument. The ITC result file is then imported by the Bia2iTC (beta release) software, enabling a comparison of the results in an Excel sheet. The combined data allows also a direct examination of possible correlations between the rate constants, affinities and the directly measured thermodynamic entities, and comparative plots can be done using Excel functionalities.
