Define protein characteristics to select the best candidates

Clearly defining interaction characteristics ensures that the best therapeutic candidates are selected as early as possible in the drug discovery and development process. Biacore systems deliver high quality data on kinetics, affinity and specificity, as well as epitope mapping for antibodies.

As potential candidates are modified to meet therapeutic requirements, Biacore systems monitor changes in the kinetic and affinity profiles so that the best candidate can be selected out at every stage. For example, efficacy and stability can be increased, effector responses enhanced using bispecific antibodies, active fragments identified and produced to penetrate tumors more efficiently.

Reducing costs in biotherapeutic development 

Early kinetic screening of hybridomas with Biacore A100 provides confident selection of antibodies with therapeutically-relevant binding properties, at an early stage in the development process. Around 800 crude hyrbidoma samples can be screened in 24 hrs,  enabling efficient, effective selection of candidates with the required kinetic profiles.

A total of 384  hybridoma samples supplied by Abgenix Inc. were screened in order to select antibodies with the required antigen-binding properties. mAbs were captured from crude hybridoma samples using an immobilized anti-IgG antibody and analyzed for binding to a single concentration of antigen.

Predicting in vivo behavior

The extremely stable temperature control of Biacore T100 makes this system well suited for studying interactions at physiological temperature, optimizing the predictive value of the binding data obtained for downstream in vivo testing and clinical use.

Deciding on dose and administration efficacy for protein therapeutics

A challenge for those working wth protein therapeutics is to minimize the dose and administration frequency in order, not only to reduce the risk of side effects and develop simple dosing regimes, but also to keep down the cost of goods.

In immunotherapeutic development for example, Biacore systems monitor immune responses in relation to time, adjuvant and dosing regime. Rapid kinetic profiling of responses gives qualitative assessment of the antibody stability in relation to the immunization regimes.

For these types of study Biacore assays have been shown to offer higher quality data when compared to traditional techniques such as ELISA . A Biacore assay is more reproducible, has a higher information content (kinetic quality assessment) and can detect immune responses earlier due to reliable measurement of low-medium affinity antibodies.

Dedicated for concentration measurements

Biacore C is a system dedicated for rapid concentration measurements required during preclinical and clinical development, manufacture and quality control of a protein therapeutic. Results are used, for example, to check process control and for batch release assays.
