Improve analysis of complex samples

Minimize non-specific binding with NSB Reducer

Components in complex sample matrices such as plasma, serum or cell lysates may bind non-specifically to the dextran surface of sensor chips, complicating the analysis of specific binding interactions. Such effects can be minimized by using NSB Reducer.

The carboxymethyl dextran in NSB Reducer has a similar structure to the dextran matrix on the sensor surface, thereby reducing  non-specific binding of complex sample components to the surface via a competition effect. Adding NSB reducer to complex samples prior to injection therefore reduces non-specific binding levels without affecting the measurement of specific binding interactions.

NSB Reducer in action

In the example shown below, ten different human plasma samples (plus a mixture of them) were injected over the carboxymethyl dextran surface of Sensor Chip CM5. The injection contact time was extended somewhat in order to increase non-specific binding levels for optimal assessment of NSB Reducer.  Levels of non-specific binding were compared with and without the addition of NSB Reducer.

Reduction of non-specific binding of plasma samples to a CM dextran surface. Despite a wide range of response levels among the plasma samples (150 to 2548 RU), NSB Reducer significantly lowered non-specific binding in all cases. The level of reduction observed was between 69-98%.
