Simplifies your interaction studies and extends your experimental range

Biotin CAPture Kit  enables reversible capture of biotinylated molecules and standardized regeneration for interaction studies in Biacore systems, offering significant advantages when setting up new assays:

Speed and convenience. Study interactions without the need to know or establish immobilization methods or regeneration protocols. Only biotinylation of the ligand is required.

Extended experimental range. Study unstable ligands or ligands for which no regeneration can be established. This is possible since the sensor chip surface is regenerated and rebuilt in each cycle using a standardized protocol.

Affordability. Save costs, time and effort by retaining the same sensor chip when you need to change ligands between experiments.

Biotin CAPture Kit provides the sensor surface, reagents and protocol for convenient, reversible capture of biotinylated ligands for interaction studies in Biacore systems. The kit comes in two versions, one for each Biacore sensor chip format. It is the ideal choice when:

• There is little time available to develop immobilization/ 
  regeneration conditions for your ligand
• Regeneration of the biotinylated ligand is difficult or
  impossible to find
• Frequent changes of biotinylated ligands are required

Technical concept:

Sensor Chip CAP is built on a carboxymethylated dextran matrix, pre-immobilized with a ssDNA oligo

Biotin CAPture Reagent consists of streptavidin conjugated with the complementary ss-DNA oligo

Biotin CAPture Reagent is injected and hybridized to the surface, followed by injection and capture of the biotinylated ligand and subsequent injection of the analyte sample.

Standardized regeneration of sensor surface is achived by disruption of hybridized DNA oligos ready for the next cycle

Application example: Studying  the interaction of TNF-alpha with Fab fragments using Biotin CAPture Kit

Biotinylated TNF-alpha was captured and the binding of antibody Fab fragments was monitored in single cycle kinetics format. The challenge in studying TNF-alpha is that it is a non-covalent trimer, which falls apart if one tries to regenerate the surface. By using Biotin CAPture Kit it was possible to capture a new aliquot of TNF-alpha to test each Fab, providing more consistent results.

The results demonstrate that Biotin CAPture Kit allows for ranking of Fab fragment binding to TNF-alpha, a ligand that is difficult to study using alternative assay formats due to its instability during regeneration.
