Application example: Studying the interaction of TNF-alpha with Fab fragments using Biotin CAPture Kit
Biotinylated TNF-alpha was captured and the binding of antibody Fab fragments was monitored in single cycle kinetics format. The challenge in studying TNF-alpha is that it is a non-covalent trimer, which falls apart if one tries to regenerate the surface. By using Biotin CAPture Kit it was possible to capture a new aliquot of TNF-alpha to test each Fab, providing more consistent results. (Data courtesy of Dr. David Myszka, University of Utah, USA)

The results demonstrate that Biotin CAPture Kit allows for ranking of Fab fragment binding to TNF-alpha, a ligand that is difficult to study using alternative assay formats due to its instability during regeneration.
