Assay design for analysis of enzyme inhibitor function using Biacore 3000 & MS

Peptide substrate (CGRP) was incubated with Trypsin immobilized on the sensor surface in the absence (step 1) or presence (step 2) of the trypsin inhibitor, aprotinin. Digested & non-digested substrate peptides were recovered from the surface and analyzed by MALDI-MS to assess inhibitor function
