Putting Biacore X100 to the test

Biacore X100 is designed to enable new users to run successful assays from day one. PhD student and protein interaction analysis novice, Thuy Tran, put this theory to the test.
Research goal:
Compare binding of several Affibody® molecules to HER2

Determine binding specificity, affinity and on/off rates

Specificity of Affibody molecules rapidly defined
Thuy rapidly demonstrated binding specificity for all Affibody molecules

Compared binding levels to immobilized HER2 and a reference protein

Affibodies (Affibody AB, Sweden) are small highly target-specific binding proteins (7 kDa). HER2 is an EGFR familiy member, and is the therapeutic target of the anti-breast cancer humanized mAb, Herceptin

Straightforward analysis of  high affinity binders using single cycle kinetics

Standard kinetic assays analyze a different concentration of sample  in successive cycles and any remaining bound sample must be completely removed by regeneration of the surface

Good regeneration conditions could not be found for the very high affinity variants, but Thuy used regeneration-free, single-cycle kinetics to easily and quickly acquire the data

Biacore X100 in commercial biotechnology

Mercodia AB (Uppsala, Sweden) provide high quality immunoassay kits (ELISAs) for clinical and research use. These are used in human and other mammalian models in the fields of cardiovascular disease, obesity and diabetes.

Label-free characterization of antibody-antigen interactions provides important, high quality data  for decision-making during product development and production in this type of biotechnology environment.
